Dimethylsulfoxide as carrier in enzyme cytochemistry.

Addition of dimethylsulfoxide (DMSO) to the incubation medium of succinate dehydrogenase in a concentration of 10% enhances the staining reaction in the hyphae of the fungus Cercosporella herpotrichoides after an incubation period of 15 min. Controls without DMSO remain unstained. DMSO causes a rapid penetration of the components of the medium through the mucilage that covers the hyphae.

The Parasitic Colonization of the Horny Layer in Tinea Versicolor

Observations of vertical sections and of tape strippings of areas infested with tinea versicolor indicate that because of their dimorphic state, these parasites can colonize the horny layer. Mycelial hyphae progress downwards into the intercellular spaces of the horny cells where they feed on nonkeratinous substances and extrude proliferating yeast cells which form clusters in nest-like cavities. The yeast-mycelial conversion gives rise to new mycelial hyphae and the cycle is complete. The parasites are actually lodged in tunnels and nests hollowed out in the horny layer. Growth and macromolecular content of the dimorphic fungus Aureobasidium pullulans and the effect of hydroxyurea and other inhibitors

The growth kinetics and the macromolecular content of the yeast and ethanol-induced hyphal forms of Aureobasidium pullulans were studied. During the morphological transition from yeasts to hyphae, both the protein and RNA content decreased significantly, the mycelial form containing only 76% of the amount of protein in the yeasts, and 38% of the RNA. The DNA was the only component tested whose level increased during the transition. Among several compounds inhibiting macromolecular synthesis, only hydroxyurea showed a remarkable effect on the morphology of A. pullulans, inducing the mycelial morphology. Relationship between ethanol tolerance, lipid composition and plasma membrane fluidity inSaccharomyces cerevisiaeandKloeckera apiculata

The lipid composition of a strain of each of two yeasts, Saccharomyces csrevisiae and Kloeckera apiculata, with different ethanol tolerances, was determined for cells grown with or without added ethanol. An increase in the proportion of ergosterol, unsaturated fatty acid levels and the maintenance of phospholipid biosynthesis seemed to be responsible for ethanol tolerance. The association of ethanol tolerance of yeast cells with plasma membrane fluidity, measured by fluorescence anisotropy, is discussed. We propose that an increase in plasma membrane fluidity may be correlated with a decrease in the sterol: phospholipid and sterol: protein ratios and an increase in unsaturation index.

Oxacyclododecindione, a Novel Inhibitor of IL-4 Signaling from Exserohilum rostratum

In a screening program for new metabolites from fungi inhibiting the IL-4 mediated signal transduction, a novel chlorinated macrocyclic lactone, designated as oxacyclododecindione, was isolated from fermentations of the imperfect fungus Exserohilum rostratum. The structure was determined by a combination of spectroscopic techniques. Oxacyclododecindione inhibits the IL-4 induced expression of the reporter gene secreted alkaline phosphatase (SEAP) in transiently transfected HepG2 cells with IC50 values of 20-25 ng/ml (54-67.5 nM). A temperature-type model for describing the relationship between fungal growth and water activity

Growth of Penicillium chrysogenum, Aspergillus flavus, Cladosporium cladosporioides and Alternaria alternata at their respective optimum temperatures was studied in Potato Dextrose Agar (PDA) medium at different water activities (a(w)) adjusted with glycerol.
